The Biologics Discovery group within the Therapeutics Discovery (TD) organization of Janssen R&D is engaged in the discovery and optimization of novel antibody and antibody based molecular entities in partnership with all therapeutic areas of Janssen. In this role you will learn about the exciting science Janssen is exploring in protein engineering that will feed into antibody and other therapeutic modalities. This position offers a phenomenal opportunity for you to engage in the discovery of next generation Biotherapeutics to address unmet medical needs as part of an outstanding team of scientists. As a member of the Protein Optimization team, you will be responsible for antibody design and optimization with a strong focus on affinity, developability and other critical attributes using structure and modeling-based design, display technologies and other emerging computational methods for protein property assessment and design. The group will also be responsible to implement high throughput screening and/or selection for the optimized antibody/protein variants to support the development of biologics such as multispecific antibodies, CAR-T/NKs, ADCs in various disease areas. Key Responsibilities


  • Train new hires and mentor other scientists, contractors, and co-op students
  • Design and perform on display-based technologies for antibody optimization, with an emphasis on phage and yeast display techniques.
  • Manage laboratory operations including such as reviewing of experimental protocols and equipment manuals, writing protocols, ordering of reagents and supplies, inventory management, instrument QC, scheduling of equipment maintenance, safety inspections
  • Design and clone antibody and protein based molecular entities
  • Perform and expand established assays as well as develop and optimize new assays. Responsibilities will include cloning, plasmid scale-up, plasmid QC, cell culture, reagent preparation, E. coli and mammalian cell culture, ELISA and other binding assays and flow cytometry.
  • Support the broad protein engineering efforts in the modality agnostic Therapeutics Discovery organization
  • A minimum of 3 years of relevant laboratory experience is required
  • Be able to independently process data, interpret results, update the database, and present results to applicable functional groups.
  • MSc or PhD or equivalent in Molecular Biology, Biochemistry, Biotechnology, Biomedical Engineering, structural biology or related field is required
Experience and Skills:


Required


  • Strong written and oral communication skills are required
  • In-depth understanding and experience of antibody engineering is required
  • Proficient knowledge of cellular/molecular biology, common biology laboratory assays such as ELISA, Luminex, and biolayer interferometry (Octet) and other assays are required
  • Demonstrated experience working in a cross-functional organization is required
  • Innovative approach and ready to step out of the box for creating and exploring new ideas with calculated risk taking is required
Preferred:


  • Background in managing incoming requests, conducting design and inventory registration of plasmids in a central database system is preferred
  • Experience in molecular biology techniques including bi-, multi-specifics, scFv, and/or chimeric antigen receptor (CAR) construct design and cloning is preferred
  • Maintain accurate inventory of generated plasmids, lab notebooks, and other records and writing of technical reports, summaries, protocols, and manuscripts are preferred
  • Experience with protein production using transient transfection/stable cell line generation using CHO and HEK-293 is preferred
  • Experience in display-based technologies is preferred
