Job Description


The Broad Institute of MIT and Harvard is looking for an exceptional candidate for a Research Scientist I to join the MIDAS project team within the Golub lab. This position affords the exciting opportunity to be part of an expanding research effort to delineate cancer dependencies in diverse in vivo microenvironments with a view towards the discovery of novel or unappreciated therapeutic targets. The candidate will assist with increasing the scale of our existing high-throughput in vivo assays using xenograft models of human cancer cell lines and next-generation cancer models. The individual will use modern functional genomics tools including emerging CRISPR genome editing technologies, next-generation sequencing, single cell RNAseq, and spatial transcriptomics to analyze the mechanism of cancer dependencies within the tumor microenvironment. The candidate will be part of a multi-disciplinary team that develops and evaluates new technologies to navigate the complexity of cancer biology revealed through our in vivo models.


The candidate is expected to work independently on a daily basis, and will be mentored by the principal investigators and senior scientists of the group. They will have the opportunity to widen their vision about cancer research by interacting with multiple collaborating laboratories and platforms at the Broad. The Broad Institute provides a vibrant and highly collaborative research environment with close links to top academic institutions across the Boston Area, providing the potential for contributions to be utilized and recognized worldwide.


CHARACTERISTIC DUTIES


  • Handle large-scale mammalian cell culture, ranging from historical cancer cell lines to patient-derived cancer organoids.
  • Routine animal handling, bioluminescent imaging, and surgical procedures.
  • Learn and execute an existing pipeline of in vitro assays and develop new in vitro assays to reconstitute the dependencies seen within the tumor microenvironment.
  • Learn, execute and further develop protocols and explore novel genomics technologies.
  • Attend team meetings, present results to collaborators, plan projects and experiments.
  • Document, compile, and analyze experimental data.
  • Prepare, order, and maintain stocks of necessary reagents, solutions and supplies.
  • Operate and maintain laboratory equipment to complete investigations.
  • Work closely in collaboration with computational scientists, Broad platforms, DFCI facilities, and other collaborators.
  • Learn and execute in vivo CRISPR screens with tumor and metastasis xenograft models.
  • Isolation of monocytes from human peripheral blood and differentiation into macrophages for co-culture experiments.

  • Strong scientific standing and background knowledge.
  • Proactive in identifying problems and solutions to address them.
  • Candidates who have a familiarity with Python or R programming languages and experience with cloud computing / cloud data management will be preferred.
  • Great attention to detail.


Qualifications


  • PhD degree in Cancer Biology, Cell Biology or related fields is required.
  • Experience and willingness to work with animals is strongly preferred, with experience generating humanized mice highly desirable.
  • Extensive experience with cell culture, molecular biology, recombinant DNA and protein biochemistry is required.
  • Familiarity with histology, hyper-plexed fluorescent microscopy, spatial transcriptomics, or flow cytometry is desirable.
  • Strong publication record and diverse research skills is required.
  • Experience with isolation and differentiation of primary human immune progenitors is desirable.
