The Gene Therapy group of Janssen Therapeutics Discovery offers an exciting opportunity to help develop novel Gene Therapies for unmet medical needs. We are investigating the use of Adeno-Associated Virus (AAV) vectors for gene replacement and to deliver therapeutic proteins to patients for disease prevention as well as treatment. The successful candidate will play a major role in helping design the best possible AAV capsids and therapeutic transgenes for gene therapy discovery efforts in retinal, neuro, and infectious diseases. The scientist will add to our capabilities for assessing optimal cargo configurations with a focus on protein biochemistry and analytics after expression in relevant cells and tissues.

Key Responsibilities

• Designing and implementing assays to analyze secreted protein products for biochemical properties, including binding, kinetic, biophysical, and structural assays.
• Designing and optimizing AAV capsids and the transgene protein product for optimal expression in the target tissue and/or specific cell type.
• M.S. with at least 4 years OR a B.S./B.A. with at least 10 years of proven experience in the pharmaceutical industry or academic environment is required
• Test engineered designs in cell culture and AAV vectors in animal models in collaboration with other Gene Therapy functional groups and therapeutic area scientists to measure gene expression levels, tissue specificity, and/or function to develop lead candidates.
• Development of novel designs that improve the stability, tissue expression, and distribution of therapeutic proteins expressed by AAV transgenes.

Experience and Skills:

Required

• Performed protein expression, purification, and characterization for at least 3 years is required
• Demonstrated ability to work collaboratively in cross-functional teams is required
• Strong critical and scientific skills allowing the individual to work independently is required
• Hands-on experience with molecular cloning and bacterial cultures for a minimum of 2 years is required
• Experience designing expression vectors for proteins and/or antibodies is required

Preferred:

• Proficient at protein purification chromatographies such as Protein A, ion exchange, and size exclusion is preferred
• Experience engineering bi- and multi-specific proteins and antibodies is preferred
• Ability to characterize protein properties and biophysics using techniques like protein titer, SEC-MALS, nano-DSF, and mass spec is preferred
• Bioinformatics expertise necessary to analyze protein sequences is preferred

The base pay range for this position is $102,000 - $163,300.

The Company maintains highly competitive, performance-based compensation programs. Under current guidelines, this position is eligible for an annual performance bonus in accordance with the terms of the applicable plan. The annual performance bonus is a cash bonus intended to provide an incentive to achieve annual targeted results by rewarding for individual and the corporation’s performance over a calendar/ performance year. Bonuses are awarded at the Company’s discretion on an individual basis.
