OBJECTIVES/PURPOSE

The Cell Engager team of the Oncology DDU is seeking a highly motivated Research Scientist II with demonstrated skills in interrogating immune and tumor cell function both in vitro and exvivo to discover and develop novel therapeutics that mobilize the immune system against cancer. As a key member of the team, the candidate will be responsible for assessing therapeutic concepts and immune engager molecules and advancing novel research molecues towards development candidates and IND. The position is hands-on and requires effective communication and documentation of results and collaboration with key stakeholders in ODDU (IOI, C2H), Global Biologics, DSRE, DMPK, Translational teams. Ideal applicant will be team-oriented, possess excellent communication and organizational skills.

ACCOUNTABILITIES

  • Evaluate the dual antigen targeting strategies in Immunotherapies as part of novel COBRA design
  • Interprets results of complex experiments
  • Will contribute to strategic discussions and goal setting with team
  • Prepares and presents comprehensive technical or project reports and formal presentations / posters to full range of internal/external audiences
  • Design and execute in vitro and ex vivo functional assays to assess therapeutic concepts aligned with principles of the COBRA and othe CE platorms
  • Executes experiments with minimum supervision and delivers comprehensive high quality data sets to enable decision making
  • Work in close collaboration with translational team to employ fluorescent-based phenotyping of immune cells in tumor FFPE tissue sections for ex vivo assessments
  • Contributes to new ideas and concept generation aligned with Cell Engager Strategy
  • Establish and validate next generation 3D tumor organoid models for ex vivo preclinical assessments
  • Support target validation efforts by characterizing receptor internalization kinetics by quantitative imaging technology
  • Maintain laboratory notebooks and keep in compliance with Takeda policies
  • Utilizes relevant information, technology and resources, contributing to complex team problem resolution and objectives
  • Identifies, plans and executes well defined scientific projects
  • Design, lead, execute T cell dependent cytotoxicity, cytokine release, mRNA profiling and other in vitro cell-based assays to support candidate lead selection process
  • Initiates investigation of new experimental approaches to specific problems using scientific theory and technical literature to fill key gaps in critical path
  • Uses defined practices and theoretical knowledge to perform analysis, interpret results and determine appropriate action
  • May serve as a PTL over time
  • Identify state of the art experimental protocols and releavnat IO assays
  • Interface with in vivo, Global biologics, DMPK,DSRE, and ITT groups to advance lead selection process build synergies across concepts
  • Build relationships with external research collaborators and CROs for cell based and translational assays
  • Utilize immune readouts such FACS based immunophenotyping, T cell activation assays, cytokine profiling that can aid with translational biomarker strategy

  • Receives high level instructions on work; determines methods on new assignments; demonstrated greater independence; may manage interns
  • Demonstrates theoretical knowledge of cell engager platforms
  • Demonstrates in-depth technical knowledge within various assay development platforms
  • Scientifically independent
  • Participates in and supervises external interactions and collaborations with companies and academic research groups and potential collaborator
  • Indepently investigates new immunological techniques, that can be applied to team projects

EDUCATION, BEHAVIOURAL COMPETENCIES AND SKILLS:

  • Experience with mammalian cell culture with best practices in tissue culture including primary human cells and tissues is strongly is required
  • Hands –on experience and ability to work with minimum supervision in carrying out high throughput data acquisition, analysis, storage and reporting by FACS CANTO, Tyto, FACS ARIA, ELISA, Luminex, MSD, IncuCyte, Nanolive and other flow based cytokine assays is requiredExperience in the design and construction of mammalian expression vectors and cell transfection and/or viral-mediated cell transduction, preferably of primary immune cells is a must
  • Ability to run, nanostring, qPCR assays and analysis is required
  • Extensive hands-on experience and knowledge in a wide range of immunological assays using cell lines and primary immune cells including T cells, NK cells, Macrophages and dendritic cells to design new approaches for immune engaging mechanisms with a focus on biologics discovery and development is expected.
  • Experience with flourescence imaging is desired

  • Experience with biologics is a must (Antibodies, ADCs, Cell therapeis etc)
  • PhD in Immunology, Cell Biology and/ or Biochemistry, or MS with 6+ years experience (industry experience preferred), or BS +8 years in Immunology, Cell Biology, Pharmacology.

Base Salary Range: 130,200.00 to 186,000.00 Employees may also be eligible for Short Term and Long-Term Incentive benefits. Employees are eligible to participate in Medical, Dental, Vision, Life Insurance, 401(k), Charitable Contribution Match, Holidays, Personal Days & Vacation, Tuition Reimbursement Program and Paid Volunteer Time Off.
